HMPM是來自生薑(Zingiber officinale Roscoe)的新穎天然物,其生物活性尚未被深入探討。乳癌為女性癌症之首。因此,本論文主旨在於,評估並比較HMPM對乳癌細胞與正常乳房細胞的生長抑制作用與機制。在MTS實驗測試中發現,HMPM具有選擇性殺死乳癌細胞(HCC1937與HS578T)的能力,對正常乳腺細胞(M10)影響較少。乳癌細胞的IC50 濃度為(70.23跟22.7 μg/ml),而正常乳腺細胞IC50 濃度為(>100 μg/ml)。在ATP 實驗中,加入氧化壓力的抑制劑N-acetylcysteine (NAC),發現加入NAC後的HMPM處理乳癌細胞的細胞存活率有提升,證實HMPM藉由氧化壓力抑制乳癌細胞生長。後續實驗透過流式細胞分析,證實HMPM影響細胞週期的改變,並誘導乳癌細胞凋亡的發生,提高活性氧化物(reactive oxygen species; ROS)的產生,且降低粒線體膜電位(mitochondrial membrane potential; MMP),最後發現HMPM會造成DNA的氧化性損傷(8-hydroxy-2-deoxyguanosine; 8-OHdG)。因此,HMPM在對三陰性乳癌細胞的抑制細胞生長與氧化壓力的上升,所引發的細胞週期的影響、粒線體膜電位的降低和DNA損傷有相關。因此,HMPM具有進一步開發作為抗乳癌物質的潛力。

HMPM is a novel natural compound derived from Zingiber officinale Roscoe. However, the biological activity of HMPM remained unknown. Breast cancer is the most crucial health issue for women cancer. Therefore, the primary purpose of this thesis is to evaluate the growth inhibition effects and mechanisms of HMPM between breast cancer cells and normal cells. The MTS test found that HMPM could selectively kill breast cancer cells (HCC1937 and HS578T) rather than normal breast cells (M10). The IC50 concentration of HMPM in breast cancer cells (HCC1937 and HS578T) were 70.23 and 22.7 μg/ml, while normal breast cells own higher IC50 concentration (>100 μg/ml). During the ATP examination, N-acetylcysteine (NAC), an inhibitor of oxidative stress, was found to increase the cell survival rate after adding NAC, confirming that HMPM inhibits the growth of cancer cells through oxidative stress. Subsequent flow cytometry experiments figured out that HMPM affects cell cycle changes, induces the tendency of breast cancer cell apoptosis, increases the production of reactive oxygen species (ROS), reduces the mitochondrial membrane potential (mitochondrial membrane potential; MMP), and finally, HMPM was found to cause DNA oxidative damage (8-hydroxy-2-deoxyguanosine; 8-OHdG). Therefore, HMPM inhibition of cancer cell growth in triple-negative breast cancer cells is related to rising oxidative stress, cell cycle effects, reduction of mitochondrial potential, and DNA damage. HMPM warrants further investigation as anticancer material.
